Protocol notes
  • March 5th PCR
  • taq polymerase use was New England Biolabs
  • annealing temperatures for arthropod reaction was 49 celsius, for denaturing was 94 celcuis, and extension 68 celcius
  • March 26th PCR
  • taq polymerase used New England Biolobs
  • Duplex PCR we used both Wolbachia and Arthropod primers
  • DNA ladder used New England Biolabs
  • annealing temperatures for arthropod reaction was 49 celsius

Results

Wolbachia presence Unknown
Confidence level Medium
Explanation of confidence level

It is unknown if my arthropod will have Wolbachia, Our controls looked good our problem is that our arthropod DNA doesn’t seem to be there, and next time we would need to add more DNA to get a more conclusive result.

Wolbachia 16S sequence
Arthropod COI sequence
