Arthropod PCR did not produce a band. Wolbachia band was faint but visible at ~430 bp. Standard lab protocol followed for all other steps.

Explanation of confidence level

Choosing Low because the Wolbachia gel band was faint rather than strong. Although the BLAST match returned 100% identity to Wolbachia endosymbiont of Armadillidium vulgare (E-value 3e-95), the weak band intensity suggests low template DNA, which lowers confidence in whether the sequence reliably represents Wolbachia infection in this specific moth specimen. The low 260/230 ratio (1.39) from the nanodrop also indicated contamination that may have affected PCR efficiency.
