Observations | The Asian lady bettle (hamornia axyridis) observed was around 5-7mm in size with a dome shaped body and varing orange to red colors, marked with just 2 balck spots on sideways mostly spotless. It was found in the tree in my school by birch hall science building .The beetle moved slowly and stayed in the leaves for long time, displaying defensive behavior by secreting a yellow liquid when distrubed. |

Protocol notes | For gel electrophoresis: Prepare Agarose gel: dissolve agarose in TAE buffer heat, pour into a casting tray and let it soldify, mix DNA/protein samples with loading buffer and add a molecular marker.Place the gel in the electrophoresis chamber ,load the samples and run at 100-150V for 30-60 minutes,Stain with ethidium bromide or coomassive brillant blue and view under UC light, for results compare the bands to the molecular marker to estimate fragment sizes. |

Wolbachia presence | Unknown |
Confidence level | Low |
Explanation of confidence level | The confidence level for obtaining DNA from the insect sample is low due to the absence of detectable DNA.Possible reasons include insufficient tissue sample, degardation of DNA,or ineffective extraction methods. These factors likely contributed to the lack of amplification, leading to an inability to observe expected results. As a result, the reliabilty of the findings is compromised and further optimization or repetition of the DNA extraction process may be required. |
