For DNA extraction we used the insect adaptation of the protocol.

March 4th, 2025:

Single PCR with Arthropod primers.

Taq Polymerase used: New England Biolabs OneTaq.

Annealing temperature for PCR reaction: 49 C.

March 25th, 2025:

Duplex PCR with Arthropod and Wolbachia primers.

Taq polymerase used: NEB OneTaq Hot Start Quick-Load 2x MM w/ Stand Buffer (M0244S).

Annealing temperature for PCR reaction: 49 C.

April 11, 2025:

Running gel electrophoresis once again to look at results of March 25th’s Duplex PCR.

Taq Polymerase used: New England Biolabs OneTaq Hot Start Quick-Load 2x MM w/ Standard Buffer (product # M0488S).

DNA Ladder used: New England Biolabs 1 kb Plus DNA Ladder for Safe Stains (product # N0559S).

I am not confident with my results because there are no clear bands in our samples (MRH6 and ELMO8). There were smears, which indicates that the DNA concentration will need to be reevaluated.
