| Protocol notes | For the DNA extraction we used the insect adaptation of the protocol On March 4th 2025 we did a single PCR with Arthropod primers which used: Taq polymerase used: New England Bio Labs One Taq Annealing temperatures for PCR reaction: 49 C On March 25th 2025 we did a duplex PCR with Arthropod and Wolbachia primers which used: Taq polymerase: NEB OneTaq Hot Star Quick-Load 2xMM w/ Stand. Buffer (M0488S) Annealing temperatures for PCR reaction: 49 C On April 11th we are running the gel of the results of March 25th using: Taq Polymerase: New England Biolabs OneTaq Hot Start Quick-Load 2x MM w/ Standard Buffer (product #M0488S) DNA Ladder used: New England Biolabs 1 kb Plus DNA Ladder for Safe Stains (product #N0559S) |
