Protocol notes | For DNA extraction we used the insect adaptation of the protocol. March 4th, 2025: Single PCR with Arthropod primers. Taq Polymerase used: New England Biolabs OneTaq. Annealing temperature for PCR reaction: 49 C. March 25th, 2025: Duplex PCR with Arthropod and Wolbachia primers. Taq polymerase used: NEB OneTaq Hot Start Quick-Load 2x MM w/ Stand Buffer (M0244S). Annealing temperature for PCR reaction: 49 C.
