| Protocol notes | For DNA extraction we used the insect adaptation of the protocol. March 4th, 2025: Single PCR with Arthropod primers. Taq Polymerase used: New England Biolabs OneTaq. Annealing temperature for PCR reaction: 49 C. March 25th, 2025: Duplex PCR with Arthropod and Wolbachia primers. Taq polymerase used: NEB OneTaq Hot Start Quick-Load 2x MM w/ Stand Buffer (M0244S). Annealing temperature for PCR reaction: 49 C. April 11, 2025: Running gel electrophoresis once again to look at results of March 25th’s Duplex PCR. Taq Polymerase used: New England Biolabs OneTaq Hot Start Quick-Load 2x MM w/ Standard Buffer (product # M0488S). DNA Ladder used: New England Biolabs 1 kb Plus DNA Ladder for Safe Stains (product # N0559S).
