Observations

We found this roly poly under some bark and leaves. It was collected in the late morning, between 50-60 degrees F. Many different insects and more roly polies were present but we only collected one.

Putative identification Arthropoda Crustacea Malacostraca Isopoda Armadillidae Armadillidium Armadillidium vulgare

Methods

Protocol notes

DNA Extraction: The arthropod was crushed thoroughly to ensure proper extraction of DNA.

Gel Electrophoresis Lanes for Arthropod Gel:

1 – PCR molecular ruler (ladder)

2 – Black-Bodies Fruit Fly

3 – Roly Poly

4 – Grass Spider

5 – Long-Legged Fly

6 – (+) Arthropod Control

7 – (-) Arthropod Control

8 – (+) DNA Control

Analysis: Our controls worked confirming our trial worked correctly and we had bands confirming that our insect samples are arthropods.

Gel Electrophoresis Lanes for Wolbachia Gel:

1 – PCR molecular ruler (ladder)

2 – Black-Bodies Fruit Fly

3 – Roly Poly

4 – Grass Spider

5 – Long-Legged Fly

6 – (+) Arthropod Control

7 – (-) Arthropod Control

8 – (+) DNA Control

Analysis: Our did not work meaning our trial was not successful and there were no bands on our gel except for our ladder meaning that our run is inconclusive most likely due to Wolbachia primers not working correctly.

Results

Wolbachia presence No
Confidence level Low
Explanation of confidence level

There was no band for this insect on our wolbachia gel but (+) Arthropod Control and (+) DNA Control did not work as expected. With no bands from these controls I am not confident in our results regarding the presence of Wolbachia in our insect.

Wolbachia 16S sequence
Arthropod COI sequence
Summary The Armadillidium vulgare was found to be negative for Wolbachia.
