| Protocol notes | We used a DNA extraction kit based on the insect adaptation of New England Biolab’s Monarch Spin gDNA Extraction kit. (Product t3010) The specimen was incubated in a water bath at 56 degrees C for 60 minutes. Our first gel image was taken on 3/24/2026 and was run at 125 volts for 20 minutes. We used the New England Biolabs 1 kb Plus DNA Ladder for Safe Stains (product # no559s) Our second gel image was taken on 4/7/2016 and was run at 125 volts for 37 minutes. We used the New England Biolabs 1 kb Plus DNA Ladder for Safe Stains (product # no559s) |
Results |
|
| Wolbachia presence | Yes |
| Confidence level | Medium |
| Explanation of confidence level | I am reasonably confident that my sample is positive. The positive and negative controls line up and are clearly visible which makes me confident that my results are accurate. A single PCR reaction should be run for higher confidence. I am confident for both rounds of PCR. |

| Arthropod COI sequence |
|
| Summary | The Harmonia axyridis was found to be postive for Wolbachia. |
