| Explanation of confidence level | My confidence level is high in that something went wrong after reviewing the final results. Since all of the controls worked correctly and PCR was correct, there is no issue there. In both the wolbachia and arthropod gels a band in lane three did not show up. Most likely, something went wrong during the DNA extraction process and the araneae was not crushed up enough. This would explain why the DNA did not come up on the arthropod gel. Due to this issue, I have high confidence that the wolbachia results are unknown and we can not say if this araneae is positive or negative for wolbachia. |
