DNA Extraction:
During the DNA extraction I made sure to be very careful. I crushed up the partial abdomen and stayed away from the exoskeleton. There was some trouble crushing the spider.

Gel Electrophoresis Lanes:
1. DNA Ladder
2. Hemiptera DNA
3. Araneae DNA
4. Positive Arthropod Control
5. Negative Arthropod Control
6. Positive DNA Control
7. Water

Analysis:
After reviewing my results, all of the controls worked correctly. The positive DNA control also worked on both gels showing that PCR went well. On the arthropod gel, the no band came up for the araneae. This is incorrect because it should have a band. Not only was their no band in the arthropod gel, but also in the wolbachia get in lane three. This gives inconclusive results.

Explanation of confidence level

My confidence level is high in that something went wrong after reviewing the final results. Since all of the controls worked correctly and PCR was correct, there is no issue there. In both the wolbachia and arthropod gels a band in lane three did not show up. Most likely, something went wrong during the DNA extraction process and the araneae was not crushed up enough. This would explain why the DNA did not come up on the arthropod gel. Due to this issue, I have high confidence that the wolbachia results are unknown and we can not say if this araneae is positive or negative for wolbachia.
