Observations

Sample was caught from a yeast trap in a wooded area of western Pennsylvania during late summer, about 64°, fair conditions.

DNA Extraction: My whole insect was included; I did not dissect it.
Gel electrophoresis lanes for CO1 arthropod PCR:
– Lane 1: Ladder
– Lane 2: Insect sample #1 (Ant 2)
– Lane 3: Insect sample #2 (Spotted Lanternfly)
– Lane 4: Positive fruit fly
– Lane 5: Negative fruit fly
– Lane 6: Positive DNA
– Lane 7: Negative water
Analysis: Gels ran as expected. Lane 6 was faint in Wolbachia gel, but still visible.

My confidence is high because all of my controls were present, ruling out a high chance of contamination. I did not have any issues with the protocol, so I am confident my arthropod is not infected with Wolbachia.
