| Observations | Found under rotting wood in damp soil in same colony as Isopod #1. It was caught at around 4:40 pm, and the temperature outside was 5°C.

| Protocol notes | DNA Extraction – This isopod was larger than the first. The lower half of the abdomen was removed and the exoskeleton was scored with a scalpel as well. It was crushed very well with no large pieces or major discolouration. Gel Electrophoresis – For CO1 arthropod The first two wells got damaged so we had to move over: 3: NEB NO551L Ladder 4: Isopod #1 5: Isopod #2 6: Isopod-R #1 7: Isopod-R #2 8: 9: 10: Positive Control 11: Negative Control 12: DNA Postive Control 13: Water / DNA Negative Control Wolbachia 16S 1. Water / DNA Negative Control Analysis: Our controls did not work/were contaminated, during Gel Electrophoresis, the Arthropod CO1 Band was present and the Wolbachia 16S was not present, however these results cannot be trusted to do low confidence level. There was likely an error in the DNA Extraction and PCR steps. |
