Protocol notes

DNA Extraction

Centrifuge briefly set to 10,000 rpm instead of 8,000 rpm for the first spin

PCR

Spilled a small amount of water on the table

Gel Electrophoresis

D3 sample (not this one) did not show positive; issue was unique to D3 in DNA Extraction lab

Results

Wolbachia presence No
Confidence level High
Explanation of confidence level

The protocol for this sample went on without any visible mistakes — we have a high confidence level in our gel results because positive and negative Wolbachia and Arthropod controls were included and the positive Arthropod and negative Wolbachia controls showed similar results in the gel
