Protocol notes

DNA Extraction

During first centrifuge step, centrifuge was set to 10,000 rpm instead of 8,000 rpm. Corrected quickly.

PCR

Spilled a small amount of water on the table.

Gel Electrophoresis

D3 sample (not this one) had issues with DNA extraction and thus did not appear on the gel. This issue does not appear to have affected any other samples.

Results

Wolbachia presence No
Confidence level High
Explanation of confidence level

The protocol for this sample went on without any visible mistakes — we have a high confidence level in our gel results because positive and negative Wolbachia and Arthropod controls were included and the positive Arthropod and negative Wolbachia controls showed similar results in the gel
