Protocol notes

This sample showed the DNA in the final PCR. However, in our fist PCR no results showed. The DNA was still on top and didn‘t come down, the marker worked though. This might be due to an error in the process. Since it worked the second time, there has to be an error in the PCR process and not in the DNA extraction. That might be a wrong amount of a solution or the wrong order of steps.

Results

Wolbachia presence Yes
Confidence level High
Explanation of confidence level

The line in the PCR wasn‘t very strong and rather faint. Also, in our first PCR no line showed.

However, when we got back our DNA data and turned it into fasta files to find out the sequence, this organism had perfect quality of data.

Wolbachia 16S sequence
Arthropod COI sequence
Summary The Philaenus spumarius was found to be postive for Wolbachia.
