Observations

Elaina A. lifted a non-pressure treated cut log in a raised garden and found a large ant colony. The ants immediately started to scatter and collect the eggs. She used a rock to carefully (in order to not crush the ants) scrape them in the vile filled with 90% Ethyl Alcohol.

Overall, things went well, but there are a few problems we encountered. In the DNA Extraction Bench Protocol, centrifuge #2 lost some liquid during Cell Lysis & DNA Preparation. After PCR was performed, the Wolbachia PCR tubes had different amounts. After gel electrophoresis the Wolbachia gel did not show the positive arthropod band, but the band did appear in the arthropod gel electrophoresis. We also had reason to believe the arthropod gel got punctured while putting the extracted DNA from ETC825 into the well for arthropod #1, so we skipped a lane.

Explanation of confidence level

In conclusion, ETC825 tested positive for Wolbachia, and CTE2026 did not test positive for Wolbachia. Our confidence level is medium because our arthropod gel electrophoresis results wer exactly as expected. But in our Wolbachia gel, our positive arthropod control did not have a band for Wolbachia. However, our positive DNA control did have a Wolbachia band.
