Captive / Cultivated? | Wild-caught |
Group | Edmund Burke School |
Observations | 4 wings, 6 legs, large mandibles, long antenna, hooks on back-legs, bright green and brown, cryptic coloration (resembles leaf), approximately 4-5 cm, moved by jumping, attracted to light, lacked long ovipositor (reason to believe it is a male specimen) |
Putative identification | Arthropoda Hexapoda Insecta Orthoptera Tettigoniidae |
Methods |
|
Extraction kit | DNeasy (Qiagen) |
DNA extraction location | Partial abdomen |
Single or Duplex PCR | Single Reaction |
Gel electrophoresis system | MiniOne |
Buffer | TBE |
DNA stain | GelGreen |

Protocol notes | The katydid did not test positive for arthropod DNA. This could be for several reasons, and I will redo part of the lab to see if I can extract DNA. |
Results |
|
Wolbachia presence | No |
Confidence level | Low |
Explanation of confidence level | Likely, the primers did not stick to the katydid’s DNA leading to no data during the Arthropod gel electrophoresis; however, it did not show positive for Wolbachia, which it would have if it was positive regardless of the primers sticking or not. to be safe, I put my confidence level as low because of the inconclusively. |
