Captive / Cultivated? Wild-caught
Group Pingry School
Observations

I began by finding a common fly in the wild. From here I captured the bug and placed it in a tube of ethanol. I then crushed the bug to prepare to perform DNA Extraction. Followed by PCR, Gel Electrophoresis, and PCR Purification.

Putative identification Arthropoda Hexapoda Insecta Diptera Muscidae Musca Musca domestica

Methods

Extraction kit DNeasy (Qiagen) blood and tissue kit
DNA extraction location Whole arthropod
Single or Duplex PCR Single Reaction
Gel electrophoresis system Standard electrophoresis system
Buffer TAE
DNA stain SYBR Safe
Gel images
Protocol notes

The protocol ran smoothly. All controls produced sensible results and the arthropod samples produced results similar to what is expected.

Results

Wolbachia presence Yes
Confidence level High
Explanation of confidence level

I am confident in my results because when I compared my DNA sequencing results to those of other flies on NCBI Blast, the sequences of both bugs were very similar. I also confirmed the structural analysis and verified the PCR Results for my bug.
