Protocol notes

The Taq Polymerase used was the New England Biolabs OneTaq Hot Start Quick-Load 2x MM w/ Standard Buffer (Product # M0488S). The DNA Ladder used was the New England Biolabs 1 kb DNA Ladder for Safe Stains(Product # N0559S). The date the  PCR for the Wolbachia Primer was set up was 3/27/2025 at 55°C. The Date for the Arthropod Primer was set up on 3/6/2025 at 49°C.

Gel Result Key:

1)DNA Ladder

2)Lamyctes emarginatus

3)Genus harmonia

4)Positive Arthropod Control

5)Negative Arthropod Control

6)Positive DNA Control

7)Water Control

8)DNA Ladder

Results

Wolbachia presence No
Confidence level Medium
Explanation of confidence level

For the Wolbachia gel, the DNA and Water Controls are accurate except the positive Arthropod control bar is faint. Nothing else showed on the gel showing a negative result for Wolbachia.

Wolbachia 16S sequence
Arthropod COI sequence
Summary The Lamyctes emarginatus was found to be negative for Wolbachia.
