Protocol notes | The control placing is switched compared to the protocol. For clarification the gel image is correctly labeled with the controls and my arthropod is labeled N, Control labels: W- = negative, W+ = Positive, H= water (-) control, D= DNA + Wolbachia DNA (+) Control, LD= DNA ladders, the other’s are my groupmates. |

Explanation of confidence level | My confidence level is low due to the fact that my observational results and my blast results did not match at all. My blast results stated that I extracted mite DNA from my arthropod which can only occur if my arthropod was originally infected with a species of mite. however from the observations and the photos you can clearly see that my insect is an odonata 100%. Furthermore, once I had put the blast result on a phylogeny it showed that it was closely related to the German cockroach which is odd since mites are not insects and cockroaches are. Lastly, there was no wolbakia present in my DNA. As a result of the lack of consistency between results my confidence level and my research is low. |
