| Protocol notes | DNA extraction: I took just the abdomen of my specimen and made sure to crush it well in the tube before combining it with the difference chemicals. Gel electrophoresis for CO1 arthropod PCR: 1 – DNA Ladder 2 – Spider specimen 3 – Bee specimen 4 – Wolbachia positive control 5 – Wolbachia negative control 6 – DNA control 7 – Water Analysis: All of our wells seemed to take well and show up good. However, I don’t think the positive control is showing up correctly. This may be due to a mess up in the extraction process or loss of DNA. |
Results |
|
| Wolbachia presence | No |
| Confidence level | High |
| Explanation of confidence level | During the process, the DNA ended up becoming loss or contaminated, causing the bands to not appear during electrophoresis. |
| Wolbachia 16S sequence | |
| Arthropod COI sequence |
|
| Summary | The Trochosa ruricola was found to be negative for Wolbachia. |
