Sample was collected on a rock next to a stream

Putative identification Arthropoda Insecta Hymenoptera

Methods

Extraction kit DNeasy (Qiagen) blood and tissue kit
DNA extraction location Whole arthropod
Single or Duplex PCR Single Reaction
Gel electrophoresis system MiniPCR
Buffer TBE
DNA stain SYBR Safe
Gel images
Protocol notes

I ground the entire sample for over 5 minutes

Analysis: My sample produced a band, but the negative insect control for CO1 showed a band. This indicates contamination, which may cause a false positive.

Results

Wolbachia presence No
Confidence level Medium
Explanation of confidence level

The positive Wolbachia control produced the expected band, suggesting that there was likely no error that would have resulted in a false negative. Although one of the negative controls also showed a band, indicating that a false positive may have occurred, our samples remained negative. Therefore, this issue likely did not affect the interpretation of our Wolbachia gel results.
