Observations

The Nomada sp. has two striped lines (yellow and black) on its abdomen, a thorax, and wings. There appears to be a stinger of sorts on its head as well, which was confirmed by the BLAST results of sequencing the DNA. The physical descriptions match the other insects in the genus Nomada. In the forest, it was found next to a small tree with a lot of sunlight.

Putative identification Arthropoda Hexapoda Insecta Hymenoptera Apidae Nomada

Methods

Extraction kit DNeasy (Qiagen) blood and tissue kit
DNA extraction location Abdomen

Gel electrophoresis system Standard electrophoresis system
Buffer TAE
DNA stain SYBR Safe
Gel images
Protocol notes

Extracted the DNA, Purified the DNA, elution of DNA, then did PCR on the DNA separately for the arthropod and Wolbachia, ran a gel on both the arthropod and Wolbachia data, then purified the DNA to send over for sequencing the Wolbachia and arthropod DNA

Results

Wolbachia presence Yes
Confidence level High
Explanation of confidence level

I have high confidence that my arthropod has Wolbachia because after running the gel, there was a distinct fragment that demonstrated the existence of Wolbachia. In addition, the existence of Wolbachia is confirmed by the BLAST results because the number of matching base pairs with other Wolbachia submissions was quite long (393 base pairs) out of the 413 total base pairs sequenced. Though the gel for the arthropod DNA was very faint, the BLAST results were clear and demonstrated that the arthropod was a of the genus Nomada. The base pair match to the genus was 632 out of a total of 658 base pairs sequenced. The arthropod gel for the Wolbachia+, Wolbachia-, and DNA control was slightly smeared, but the Wolbachia gel for the Wolbachia+, Wolbachia- was quite clear, depicting a dark fragment for the Wolbachia+ and not showing up for Wolbachia-.
