Observations

Caught by in an office space during late summer, 70º. Pale yellow, all 8 legs intact, body less than an inch in length.  Urban habitat.

No known mistakes made, protocol followed.

Gel electrophoresis, Arthropod CO1:

C1 was the Aracneae , C2 and C3 were other arthropods. W+ was positive for Wolbachia, W- was negative for Wolbachia but still contained arthropod DNA. Water was the control and no band appeared, confirming that the samples were not contaminated with other DNA.

Gel electrophoresis, Wolbachia:

The Aracneae was C1, C2 was another sample, and so was C3. W+ was the only lane with a band because it was the positive control. W- had no band as it was the negative control. Water also had no band which confirmed no contamination.

Explanation of confidence level

We have high confidence because none of the controls showed results that were not expected. Water had no band in any of the gels, W+ and W- both had bands for the CO1 gene, and only W+ had a band for the Wolbachia gel. As far as we know, no mistakes were made in relation to this sample.
