Observations

Caught on a bright and clear day in a pollen garden right outside the building where we have class. Caught by picking up with gloved hands and then putting into a bug net. Was caught around 1:00 PM, and was one of eight caught.

Putative identification Arthropoda Insecta Orthoptera

Methods

Extraction kit DNeasy (Qiagen)
DNA extraction location Abdomen
Single or Duplex PCR Single Reaction
Gel electrophoresis system MiniPCR
Buffer TBE
DNA stain GelGreen
Gel images
Protocol notes

DNA Extraction: I removed a majority of the insect that wasn’t its abdomen so that I could primarily focus on the innards found exclusively in the abdomen. I then took great care to crush it up thoroughly in a solution until I saw minimal/no free-floating big clumps of debris.

Analysis: My controls all worked, and while it’s faint, you can see bands for both the Arthropod DNA and the Wolbachia DNA, showing that Wolbachia can be present in Schistocerca americana.

Results

Wolbachia presence Yes
Confidence level Medium
Explanation of confidence level

I put medium as both the bands for the arthropod and Wolbachia DNA are kinda faint. Controls all worked though.

Wolbachia 16S sequence
Arthropod COI sequence
Summary The Orthoptera was found to be postive for Wolbachia.
