Protocol notes

The following lane order for our gel:

1st from left: DNA from other insect
2nd left: DNA lasius brunneus
3rd from left: positive arthropod control
4th  from left: negativ arthropod control
5th from left: gDNA
6th  from left: water
7th from left: DNA marker

Results

Wolbachia presence No
Confidence level Medium
Explanation of confidence level

All of the controls worked except the negative arthropod control.

For the first and second ones (see reference above “protocol notes”) were used 400 microliters insted of 50 microliter TE buffer for the DNA-Elution. It had no influence on the results, because they were still clearly visible.

Wolbachia 16S sequence
Arthropod COI sequence
Summary The Lasius brunneus was found to be negative for Wolbachia.
