Observations

I found this spider on the walls of my house, it had short fuzz on its body and was pretty small. It was the only spider I saw in my house and I haven’t seen another one like it before.

Putative identification Arthropoda

Methods

Extraction kit DNeasy (Qiagen) blood and tissue kit
DNA extraction location Abdomen
Single or Duplex PCR Single Reaction
Gel electrophoresis system MiniPCR
Buffer TBE
DNA stain SYBR Safe
Gel images
Protocol notes

DNA Extraction: I cut off all the legs and used DNA from the abdomen, crushed it up really well.

The negative control in arthropod DNA showed up which means something went wrong during the protocol. Majority of our data did not result in visible bands which could also be due to a lack of Wolbachia or reagents that weren’t working properly.

Results

Wolbachia presence No
Confidence level Low
Explanation of confidence level

In our experiment, the negative control for the arthropod DNA unexpectedly showed a band, which invalidated the rest of our results. Additionally, none of our actual samples showed a clear band for arthropod DNA. Even though the Wolbachia positive and negative controls worked exactly as they were supposed to, the issues with the arthropod DNA make us uncertain about our overall findings.

For the fasta and ab1 upload, I’m not uploading anything because I didn’t get any viable DNA or Wolbachia levels from the gel electrophoresis.

Wolbachia 16S sequence
Arthropod COI sequence
Summary The Arthropoda was found to be negative for Wolbachia.
