Protocol notes

We followed protocol on website.

  • The spider sample is in lane 2 of both the top and bottom rows of the gel.
  • The top row used arthropod primers while the bottom row used Wolbachia primers. In addition, the 8th well of the top and bottom rows is the DNA ladder because the 1st well of both rows didn’t have enough DNA ladder sample.

Results

Wolbachia presence Yes
Confidence level Medium
Explanation of confidence level

It is medium because Wolbachia DNA was detected in the spider sample, however, when using arthropod primers, there was no evidence from the electrophoresis that the spider was an arthropod when we know, in fact, that it is.

Wolbachia 16S sequence
Arthropod COI sequence
Summary The Insecta was found to be postive for Wolbachia.
