Protocol notes

It was difficult to put the mixture into the gel. I poked it a bit which worried me that the DNA would come out. Flipping the holder upside down for a long time gave me a similar worry. The pellet was light brown and all gathered at the bottom.

Results

Wolbachia presence Yes
Confidence level Low
Explanation of confidence level
  • I didn’t have an arthropod
  • Positive gel control didn’t show band for arthropod
  • Positive DNA showed for arthropod but had an odd double band
  • No band for Wolbachia DNA
Wolbachia 16S sequence
Arthropod COI sequence
Summary The Amphipoda was found to be postive for Wolbachia.
