Methods

Extraction kit DNeasy (Qiagen) blood and tissue kit
DNA extraction location Whole arthropod
Single or Duplex PCR Single Reaction
Gel electrophoresis system MiniPCR
Buffer TBE
DNA stain GelGreen

Results

Wolbachia presence No
Confidence level Medium
Explanation of confidence level

The bar for all of our arthropod DNA gel electrophoresis was faint, even the controls indicating that something likely went wrong with the PCR. Therefore, even though our Wolbachia electrophoresis showed up properly we still only have mid confidence given the issues with our arthropod DNA.

Wolbachia 16S sequence
Arthropod COI sequence
Summary The Philaenus spumarius was found to be negative for Wolbachia.
