Protocol notes

The ticks were stored in 90% ethanol for about three months prior to extraction.

DNA Extraction: The DNA was extracted from the abdomen. The abdomen was sliced open due to the tough exoskeleton. All of the Dneasy (Qiagen) protocols were followed. The extraction took two days to complete.

PCR: Mini One Taq polymerase was used.

Gel Electrophoresis:  Mini PCR was used. All of the lanes worked well and were bright and clear.

Results

Wolbachia presence No
Confidence level High
Explanation of confidence level

All of the controls worked and the bands were bright and clear. All of the protocols were followed exactly.

Wolbachia 16S sequence
