Buffer TBE
DNA stain GelGreen
Gel images
Protocol notes

When opening the container, (we think) dirt fell in, which is what the specks are at the top of the gel. The bar for all of the arthropods showed up, except for lane 4, so something could have gone wrong in the process for that one. All of the others were extremely light and “weak”, so we think we might have punctured the gel when inserting each sample.

Results

Wolbachia presence No
Confidence level Medium
Explanation of confidence level

We have a medium level of confidence, as our Wolbachia gel showed up pretty decently, however based on our Arthropod results, it would be unreasonable to say we have high confidence. Only the arthropod in lane three tested positive for Wolbachia. Our controls were accurate.

Wolbachia 16S sequence
Arthropod COI sequence
Summary The Hemiptera was found to be negative for Wolbachia.
