Observations

The springtail was sampled in the fall from a forest found on campus. It was found under natural cover on the forest floor.

Putative identification Arthropoda Hexapoda

Methods

Extraction kit DNeasy (Qiagen) blood + tissue kit with modification P2 for ATL + N3 for AL
DNA extraction location Whole arthropod
Single or Duplex PCR Single Reaction
Gel electrophoresis system Standard electrophoresis system
Buffer TAE
DNA stain GelRed

Protocol notes

Results

Wolbachia presence No
Confidence level High
Explanation of confidence level

The gels showed the following results:

  • The presence of bands at the expected band size for arthropod (Co1) gene at 708pb for the + and – Drosophila control samples and for the DNA control.
  • The was also the Wolbachia (16S rRNA) band present at 438bp for the +Drosophila control sample and DNA control.

Since the expected bands were present for the specific controls, we are confident that our results are valid.

Wolbachia 16S sequence
Arthropod COI sequence
Summary The Hexapoda was found to be negative for Wolbachia.
